Is there a way that Windows Server 2003 can support all the following roles
using only 1 Network Card:

File Server
Print Server
Domain Controller
DNS/DHCP Server
Applications Sever

and

VPN Server (always causes internal network to loose connectivity when
installed)

It is possible but it is a bad idea (unless you run sbs server which is
designed to run in that config).

The major problem is that, as soon as a VPN client connects, the server
is multihomed (because the server acquires an additional IP for its internal
interface, which is the end-point of the VPN tunnel). If you must use this
setup, look at KB292822 which outlines the problems and ways to handle them.
