I'm about to plunge into DSL and wireless networking for the first time. We
have two computers in two separate buildings about 100 feet apart. One
computer is on the second floor of its building, so the elevation is higher.
Will it be beneficial to locate the router at this computer? Does elevation
matter?

Why do you think a radio station transmitter antenna is built so high? The
higher you can get a transmitter/receiver the better the signal will carry.

It's that simple.

Not really that simple. Transmitter antenna need to be a certain
distance from the ground plane for proper operation. It's a matter of
wavelength.

In your case it would only matter to clear any obstacles.

Generally yes, but it may well depend of your local conditions, interference
and obstacles.
Get the wireless first and experiment before deciding where to place it then
connect to DSL.

To clear the trees and buildings !

Line of sight works best so 'matching' their heights would likely give best
signal - aside from beam-width if you are using antennas.
