Is there a way for me to assign to all clients a GPO that prevents users
from surfing to specific websites and denies them access? All the clients
would be using IE as their default browser.

Yes, you can use a GPO to import Restricted Sites;

User Configuration/Windows Settings/Internet Explorer
Maintenance/Security/Security Zones and Content Ratings
